The Biosynthesis of Nicotinamide Adenine Dinucleotide by Rabbit¡¯ Matured-Leukocytes in Vitro


Abstract
The studies on the biosynthesis of nicotinamide adenine dinucleotide from niacin(163 mM) or nicotinamide(163 mM) have been. investigated in the rabbit¢¥ matured-leukocytes (58x1O3/mm3). The incubation of rabbit¢¥ matured-leukocytes with nicotinic acid or nicotinamide in the presence of glucose, glutamine and ATP-containing, Krebs-Ringer phosphate buffer(pH 7.4) resulted in the uptake of the. vitamin by matured-leukocytes.
The incorporation of nicotinic acid or nicotinamide into the precursor of NAD biosynthesis, namely, nicotinic acid mononucleotide, nicotinamide mononucleotide, desamido-NAD and NAD, as analyzed by Dowex 1-formate column chromatography and by paper chromatography. The uptake of nicotinamide by the matured-leukocytes, on the other hand, had found that it was lesser than those of nicotinic acid.
From these results, it might be concluded that nicotinic acid or -nicotinamide is actively taken up by the rabbit¢¥ matured-leukocytes and that a considerable portion of the nicotinic acid or nicotinamide taken up is incorporated to NAD via a biosynthetic pathway, e.g., Preiss-Handler¢¥s pathway which is operating in the other tissues like liver and erythrocytes.
